TePixJ を酸性尿素で変性させ、スペクトル解析を行ってその発色団の同定を試みた。その結果TePixJの発色団はフィコビオロビリンである事が示唆された。フィトクロムの発色団はフィトクロモビリン、フィコシアノビリン、ビリベルジンのいずれかであるが、フィコビオロビリンは共役二重結合系がこれらの発色団より短いので、より短波長の光を吸収する事ができる。またフィコシアノビリン産生大腸菌で発現させたTePixJの発色団がフィコビオロビリンであるという結果を得た。このことからフィコビオロビリンはTePixJ自身がフィコシアノビリンを異性化することで供給されていることが示唆された。TePixJは光変換活性、発色団を結合するリアーゼ活性、異性化活性といった多機能性を保持した光受容体であることが分かった。

BLUF型フラビン結合センサーTePixDの部位特異変異解析により、フラビンの光反応に大きな影響を与える残基を同定した。Gln50をAsnに置換したタンパク質はフラビンの長波長シフトを起こさず、代わりにトリプレットが生成し、効率は低いがフラビンの光還元が引き続いて起こった。同様の現象は、Gln50と相互作用しているTyr8をPheもしくはAlaに置換した変異タンパク質でも起きた。しかし、 Gln50をAlaに置換したものではトリプレットも生成せず、光還元も長波長シフトも示さなかった。一方、Asn32をAlaに置換した変異タンパク質はFT-IR解析で明・暗ともにC(4)=Oと水素結合していることが示された。以上の結果は、Gln50のアミド側鎖がフラビンのC(4)=Oと N(5)に正しく配向することがBLUF型光反応に重要であることを示しているとともに、この配向が崩れると、フォトトロピン型の光反応に似た挙動をとりやすいことを示している。論文の図をHPトップページに追加。

シアノバクテリアの正の走光性の光受容体PixJ1をSynechocystis細胞より部分精製した。得られたPixJ1は共有結合で開環テトラピロールを結合しており、425-435 nm吸収型(Pb)と535 nm吸収型(Pg)の間で可逆的な光変換を示した。PixJ1はフィトクロムと相同性を示す2個のGAFドメインをもつが、その後半のGAFドメインの Cys-HisモチーフをAla-Alaに変えた変異体タンパク質は色素結合を示さなかった。このように、PixJ1は植物フィトクロムと似てはいるが、独自の光受容体であり、シアノバクテリアに広く分布しているので、その関連分子を含めて、「シアノバクテリオクロム」と命名した。
